In a word - no. Only via Istanbul or Ankara.

Buses easy and cheap but take time with frequent stops for toilet and food.

I would think you're probably looking at about 30 hours by bus. Dalaman-Mugla-Ankara-Van-Diyabakir, but that's a guess.
